New Substrate for Collagenase 3/MMP-13

Peptide substrates are useful for the determination of protease enzyme activity which is also an effective method for screening new potential therapeutic protease inhibitors.  A new highly sensitive substrate for assaying MMP activities is now available. Evaluation requires HPLC separation of peptide fragments (monitored at l = 350 nm). The substrate cleavage between Gly-Met is determined by HPLC separation (monitored at l = 350 nm). kcat/Km (M-1s-1) at 37°C, pH 7.5, is 4.22 x 106 for MMP-13, 3.9 x 105 for MMP-9, 5.14 x 103 for MMP-1, and 3.14 x 103 for MMP-3.  The specific substrate described below has been especially developed for the accurate determination of MMP-13 activity and will help users monitor the effectiveness and specificity of potential inhibitors.
